Recently, the reporter learned from the Kunming Institute of Zoology of the Chinese Academy of Sciences that the third-generation gene sequencing field has once again made important breakthroughs. The new technology can greatly reduce the cost of three-generation sequencing, and provides a good opportunity to promote the sequencing technology to the third-generation technology industry.
At present, the latest three generations of gene sequencing have reached 1 megabyte of ultra-long reading, which has many advantages over the second-generation technology. It is undoubtedly the future of sequencing technology, but the third-generation technology also has high error rate of sequencing base level and high sequencing cost. No disadvantage.
For the frontiers, the team led by Prof. Zhang Yaping and Prof. Ma Zhanshan from the Kunming Institute of Zoology, Chinese Academy of Sciences recently released a hybrid assembly strategy and software technology that uses 10 times genome sequencing to assist in three-generation sequencing. The researchers conducted demonstration tests using three generations of human genome sequencing data published by the University of California, Jayne et al., in Natural Biotechnology in 2018. The results show that the new method can reduce the sequencing depth from 35 times used by Jayne et al. 7 times, a reduction of 80%; converted to the cost of sequencing, the cost of the new technology is about a quarter of pure three-generation sequencing. This technology invention patent has been accepted, and related papers have recently been published online in the international journal Genomics.
Because the special labeling method adopted by the new technology can more effectively assist the assembly of the third-generation sequencing gene data, it is likely to incite the cooperation and competition between the hardware manufacturers in the current sequencing market. The release of new technologies will not only broaden the cooperative choice of gene sequencing organizations, but will also improve the advantages of other technologies they operate, especially single-cell sequencing technologies.
The peer reviewers in Genomics pointed out that the idea of ​​combining the 10x genome with the third-generation sequencing method by the new technology is quite obvious and may have been done in some sequencing centers, which may not be enough to prove Its novelty. On the other hand, the new software adds substantial value to gene sequencing research.
