High-speed refrigerated centrifuge operation precautions

The centrifuge chamber temperature of such centrifuges typically drops below zero degrees Celsius. The rotational speed can reach 10,000 rpm or more, and the performance and structure of the refrigerated centrifuge are used. The angle turret used in the high-speed refrigerating centrifuge is mostly made of titanium alloy or aluminum alloy. The centrifuge tube is a polyethylene hard plastic product with a cover. Such centrifuges are often used to collect microorganisms, cell debris, cells, large organelles, sulfate precipitates, and immunoprecipitates. So, what are the key issues that need to be addressed when operating a high-speed refrigerated centrifuge?

The main issues that need to be addressed in operating high-speed refrigerated centrifuges are:

1. The amount of solution loaded in the centrifuge tube must be appropriate. The stainless steel tube has no cover and can only be installed 2/3. The plastic tube can be attached to the shoulder. The cover must be covered and not allowed to leak. Empty tube centrifugation will deform. The use of organic solvents in plastic pipes must comply with the regulations.

2. Centrifuge tubes must be placed in pairs and must be strictly balanced with a deviation of <0.1g.

3. No idling is allowed, and the handle must be used to prevent the rotor from slipping. Turn the head to be light and steady. When you lower the handle, hold the handle with your hand and turn the head. The rotating head cover should be tightly closed, and the cover is not allowed to be centrifuged.

4. Do not open the cover during centrifugation. Do not support it on the centrifuge. If there is abnormal sound and vibration, stop immediately.

5. After using the rotor, it must be taken out from the rotor room in time and wiped dry. Wipe the hole carefully with a hole stick. If there is a solution overflow, it must be cleaned, wipe the condensate inside the rotor, and open the door to dry the rotor room.

6. The centrifuge must be reserved and must be registered after use.

7. Those who have not been trained and assessed cannot use it.

8. Choose the right rotor and speed, never use it.

9. Choose the right temperature, usually 4 ° C, do not lower than the organic solvent, so as not to freeze, damage the centrifuge tube and rotor.

10. Before using the rotor, wipe the tube hole with a hole rod and carefully check for cracks and white spots on the bottom of the hole. If so, turn around and scrap.
